SCF/c-KIT Effector Reporter Cell

I. Background

      受體(ti) 激酶在正常細胞功能中的作用至關(guan) 重要，人類有 58 種已知的受體(ti) 激 酶，該家族最重要的成員之一是 c-KIT。c-KIT 也稱為(wei) CD117，是一種由 KIT 基因編碼的 III 型受體(ti) 激酶。它以幹細胞生長因子（SCF）為(wei) 配體(ti) ，因此被稱為(wei) 幹細胞生長因子 受體(ti) （SCFR）。 c-Kit 的信號轉導通過其配體(ti) SCF 蛋白的結合而被激活，通過自磷酸化引 起 c-Kit 二聚化和激活，並啟動磷酸化級聯反應，從(cong) 而調節細胞生長。c-Kit 磷酸化可觸發 多種信號轉導通路，包括 JAK/STAT、RAS/MAP 激酶通路、PI3 激酶、PLCγ通路和 SRC 通 路。SCF/c-KIT 信號轉導通路的激活通常與(yu) 細胞增殖、遷移和存活有關(guan) ，從(cong) 而調節造血、 色素沉著和精子發生的關(guan) 鍵功能。

 
II. Description

     SCF/c-KIT Effector Reporter Cell 報告基因藥靶模型很好的模擬了體(ti) 內(nei) SCF/c-KIT 的信號轉導過程，原理見下圖所示。
