| I. Background | |
血小板衍生生長因子（PDGF）是一類重要的生長因子，主要由血小板、內(nei) 皮細胞和 巨噬細胞分泌，在細胞增殖、遷移、分化和血管生成等生物學過程中發揮關(guan) 鍵作用。它不 僅(jin) 在正常生理過程中（如傷(shang) 口愈合和組織修複）起重要作用，還在多種病理過程中（如癌 症、纖維化和動脈粥樣硬化）扮演重要角色。PDGF 家族包括 PDGF-A、PDGF-B、PDGF- C 和 PDGF-D 四種亞(ya) 型。PDGF 通過與(yu) 其受體(ti) （PDGFR）結合，激活下遊信號通路，調控 細胞行為(wei) 。 血小板衍生生長因子受 0 體(ti) α（PDGFRA），又稱為(wei) CD140A 或 PDGFR2，是一種細胞 表麵激酶受體(ti) ，屬於(yu) 血小板衍生生長因子家族。PDGFRA 與(yu) 配體(ti) PDGF 結合後會(hui) 二 聚化以及磷酸化，激活包括 JAK-STAT3、PI3K-AKT-mTOR 和 RAS-MAPK 等下遊信號通路， 在調節關(guan) 鍵細胞功能如增殖和凋亡方麵有重要作用。PDGFRA 蛋白被激活異常時，則會(hui) 導 致腫瘤的發生並促進腫瘤血管生成。 | |
